Teleost TRAF7, a protein functions in the host antiviral responses via NF-κB and IRF3/7 mediated signaling

Tumor necrosis factor receptor-associated factors (TRAFs) play vital roles in tumor receptor (TNF-R) and interleukin-1 receptor/Toll-like (IL-1R/TLR) mediated signaling pathway. However, the role that TRAF7 plays host immune responses is largely unknown comparison to extensive in-depth research has been conducted on other members of TRAF family. Notably, Lc -TRAF7, a cloned ortholog, was discovered large yellow croaker ( Larimichthys crocea ) current study, which an open reading frame (ORF) 1,962 base pairs encodes protein 653 amino acids (aa). -TRAF7 consisted RING finger domain, coiled-coil seven WD40 domains, with genomic organization 20 exons 19 introns. Luciferase assays demonstrated overexpression activation NF-κB, IRF3, IRF7, IFN1 promoters. In addition, domains pivotal NF-κB promoter activation, whereas were essential activation. could suppress SVCV proliferation EPC cells, levels IRF3 , IRF7 ISG15, ISG56 RSAD2 TNF-α up-regulated under LYCMS cells. These findings collectively implied may as important regulator antiviral via well IRF3/7 involved pathways.
